Book Description

Ion channels underlie a broad range of the most basic biological processes, from excitation and signaling to secretion and absorption. Like enzymes, they are diverse and ubiquitous macromolecular catalysts with high substrate specificity and subject to strong regulation. This fully revised and expanded Third Edition of Ion Channels of Excitable Membranes describes the known channels and their physiological functions, then develops the conceptual background needed to understand their architecture and molecular mechanisms of operation. It includes new chapters on calcium signaling, structural biology, and molecular biology and genomics. Ion Channels of Excitable Membranes begins with the classical biophysical work of Hodgkin and Huxley, continues with the roles of channels in cellular signaling, then develops the physical and molecular principles needed for explaining permeation, gating, pharmacological modification, and molecular diversity, and ends with a discussion of channel evolution. Ion Channels of Excitable Membranes is written to be accessible and interesting to life scientists and physical scientists of all kinds. It introduces all the concepts that a graduate student should be aware of but is also effective in advanced undergraduate courses. It has long been the recognized authoritative overview of this field used by all neuroscientists.

5 out of 5 stars Excellent and useful text........2006-11-17

This text is an encyclopedia of information from kinetics to structure to interactions. It's got a reserved space on my academic bookshelf.

5 out of 5 stars Destined to become an Oprah Book Club selection.......2005-12-08

Some books have opening sentences that grab you in an instant. "Call me Ishmael." "It was the best of times, it was the worst of times." And now, Hille joins the list of authors to reach out and grab us, with the awe-inspiring "Ion channels are macromolecular pores in cell membranes."

Even after my fifth reading, that line still gives me the shivers.

But it takes more than a brilliant opening to make a great book, and Hille delivers. From a distinctly jaunty derivation of the Nernst equation to the page-turning thrills of non-stationary fluctuation analysis, the book is hard to put down. It does bog down a little in Chapter 10, "Elementary Properties of Ions in Solution"--after all, is there anyone who isn't already aware of the fundamentals of electrodiffusion? But this is really a minor trifle in an otherwise masterful work.

It's just a matter of time before Oprah gives this book a nod; buy it now and avoid the rush!

5 out of 5 stars Keeps getting better with each Edition.......2002-03-27

This book is a must-have for anyone working with excitable cells, be they experimentalists or modelers. It's a great reference book; everything is clearly indexed and almost compulsively cited. I've been using this book since its first edition, and it just keeps getting better each time.

5 out of 5 stars Rara avis.......2002-02-09

This classic deserves all the praise it gets. It is an excellent introduction to the molecular biology of channels, told with an eye to history as well as science.

5 out of 5 stars A classic. Still authoritative........2001-08-13

This is an authoritative textbook on ion channels that is often quoted in numerous textbooks in neurophysiology. Hille himself has contributed a lot in this field. The Hogkin-Huxley model is the de facto standard model in neurophysiology but certain things about neurons cannot be explained only with voltage-controlled sodium and potassium channels. With the patch-clamp technique, we have found a way to explore other interesting channles. To remind you, there are a world of ion channels! Hille's textbook show there are indeed a variety of channels most of which are poorly understood. As a reviewer below has commented, it also gives a unique insight into the history of neural membrane research. Concepts are explained with clearity. Details measurement techniques.

5 out of 5 stars A truly gripping read.......2005-12-08

A frequent complaint about neuroscience books is that they are a bit dry, but I am pleased to report that "Single Channel Recording" is as taut and suspenseful a tale as has ever hit the mystery shelf. Jumping immediately into the action with Chapter one, "A Practical Guide to Patch Clamping", the book provides a sinister foreshadowing of hidden dangers yet to come. Will recording noise preclude detection of the crucial signals? Will junction potentials throw the investigation off completely? And what of poor Calcium?

I don't want to give away the explosive finale (although I will give a hint: dielectric membrane breakdown), but suffice it to say that by the time the book is over, you will be exposed to a bigger bag of tricks than was ever dreamed up by "Q branch" from the James Bond series (The analogy was obviously on the authors' minds, with direct references to the "Q matrix").

Not only is the plot crisp, but the writing is also highly satisfying and complex, sometimes with a whimsical air that vaguely recalls Vonnegut's "Breakfast of Champions" but other times with a cold analytical nature that more closely resembles Truman Capote's "In Cold Blood". The book is truly a masterpiece, and Neher and Sakmann work as well together as their Sesame Street namesakes. Dust off the shelves for another Nobel, boys, this time in literature!

5 out of 5 stars The Patcher's Bible.......2000-03-31

Single Channel Recording is the possibly the single most useful text for the patch-clamp novice, and the text continues to be a fundamental resource for veterans. Sakmann and Neher, inventors of the technique, have compiled a book to give both theoretical background and practical advice regarding both experimental setup and data analysis. Whole-cell, single channels, and perforated patch are discussed. The practicalities of suitable patch glass, noise abatement, and capacitance cancelation are also covered. This book is a must have for all electrophysiologists.

    Book Description

Ion channels are membrane proteins that act as gated pathways for the movement of ions across cell membranes. They play essential roles in the physiology of all cells. In recent years, an ever-increasing number of human and animal diseases have been found to result from defects in ion channel function. Most of these diseases arise from mutations in the genes encoding ion channel proteins, and they are now referred to as the channelopathies.
Ion Channels and Disease provides an informative and up-to-date account of our present understanding of ion channels and the molecular basis of ion channel diseases. It includes a basic introduction to the relevant aspects of molecular biology and biophysics and a brief description of the principal methods used to study channelopathies. For each channel, the relationship between its molecular structure and its functional properties is discussed and ways in which genetic mutations produce the disease phenotype are considered.
This book is intended for research workers and clinicians, as well as graduates and advanced undergraduates. The text is clear and lively and assumes little knowledge, yet it takes the reader to frontiers of what is currently known about this most exciting and medically important area of physiology.

    Key Features
    * Introduces the relevant aspects of molecular biology and biophysics
    * Describes the principal methods used to study channelopathies
    * Considers single classes of ion channels with summaries of the physiological role, subunit composition, molecular structure and chromosomal location, plus the relationship between channel structure and function
    * Looks at those diseases associated with defective channel structures and regulation, including mutations affecting channel function and to what extent this change in channel function can account for the clinical phenotype

    Customer Reviews:

5 out of 5 stars The answer is in here ...and here is the question:.......2001-05-20

The author has a splendid writing gift, and she has produced a broad and wonderfully clear survey of the state of the art in this vigorous field. For a detailed summary of what you will find in the book, check the editorial review (above) from the New England Journal of Medicine.

It seems to me the book might well be read in the context of two other important books in this field: One is Bertil Hille's classic, "Ionic Channels of Excitable Membranes." As a science gathers momentum (and this one is certainly surging) we tend to lose track of what is known and what is simply assumed. Hille's book will fill in some blanks at the fundamental level - and show you exactly where the underlying assumptions are in this science. If you are at all skeptical, and of course you should be, you will like Hille's calm precision and care.

The other background book is Spikes, by Rieke et al. The implication of Spikes is that Adrian was wrong and that, therefore, all of us have been wrong about what nerves actually do - and wrong since 1926. The authors put this rather more diplomatically than I have, but there it is: Adrian wrong.

Spikes summarize evidence accumulated since about 1993 that a single nerve impulse, all by itself, can somehow convey information to the brain. This shocking news will have to be either explained or explained away in terms of the biochemical machinery of the neuron. The current explanation (which is based on precise arrival timing) would seem to rely upon the physiological equivalent of a quartz crystal, um, a device we don't often come across in biochemistry.

It would be my guess that a better understanding of ion channels will point to a more biologically realistic solution. And a new and better picture of how the neuron works.

Ion Channels and Disease is the most current and broadest survey of the subject. The key to the problem is probably in here somewhere, or is referenced here, and is waiting to be discovered. I would pay particular attention to any type of evidence for linkage, structure or signaling between "individual" channels. Linkage between discrete trans-membrane ion channels could create a longitudinal channel running the length of the nerve, probably many of them. A multi-channel axon - a cable rather than a wire -- would be one possible solution to the new mystery of how a single impulse can be freighted with graded information.

5 out of 5 stars Fantastic Compendium of Channelopathies.......2000-01-04

This is a fantastic book for anybody interested in ion channels or molecular medicine. If you happen to be a graduate student preparing for an exam in the field - this is your salvation! Personally, I read it with enormous delight. What a great compendium of this fascinating, quickly growing field! The style is very elegant, everything is lucid, the concepts come through crystal-clear. This was certainly an enormous amount of work and the book will be helpful to many in the field - for brushing up on channels that you don't work on, for checking things quickly, for teaching, and just for fun!

5 out of 5 stars The very first book of a kind.......1999-11-11

This is really a landmark achievment to write such a good book in such a way, understandable .

5 out of 5 stars Outstanding! Essential reading in modern molecular medicine.......1999-10-18

This is a lucid discussion of the role of ion channels in disease. It begins with a wonderful introduction to molecular biology and physiology of ion channels suitable for neophyte as well as the seasoned investigator. Then individual ion and ligand gated channels are discussed in individual chapters. After the basic properties of each channel are introduced, diseases of the channels are discussed.

    Book Description

with simulations and illustrations by Richard Gray

Problem solving is an indispensable part of learning a quantitative science such as neurophysiology. This text for graduate and advanced undergraduate students in neuroscience, physiology, biophysics, and computational neuroscience provides comprehensive, mathematically sophisticated descriptions of modern principles of cellular neurophysiology. It is the only neurophysiology text that gives detailed derivations of equations, worked examples, and homework problem sets (with complete answers).

Developed from notes for the course that the authors have taught since 1983, Foundations of Cellular Neurophysiology covers cellular neurophysiology (also some material at the molecular and systems levels) from its physical and mathematical foundations in a way that is far more rigorous than other commonly used texts in this area.

    Customer Reviews:

4 out of 5 stars Mathematical Cellular Neurophysiology.......2004-06-11

This book is chock full of equations concerning the Hodgkin-Huxley Model, Kinetics of Ionic Channels, Presynaptic & Postsynaptic Transmission, LTP, LTD, quantum vesicular release and reuptake, & a "scant" neural net theory. There are a plethora of 'end of chapter' problems with derivations & applications to solve. The book is going to be confusing and tedious, even for computationally trained neuroscientists. For me, this book is a reference only. The format is similar to Thomas Weiss' Cellular Biophysics 2 Volume Set. There is too much on the Hodgkin-Huxley Model which is very important historically, but of less interest today than it was in the 1960s. The authors also use cable theory to model neuronal & dendritic function.

    2 out of 5 stars Pick a different book.......2001-10-11

This was the assigned text for a short section on electrophysiology in my first year graduate neuroscience course. I did not have a math or electronics background, but had worked in a neurophys lab for two years. I managed to learn stuff from this book, but it was hard work and I couldn't have done it without my lecturer and classmates. It's obvious that the authors really know their stuff, but being able to explain it to novices is another thing... And I agree with the other reviewer who noted that the index is *completely* useless, almost random lists of page numbers after topics...
Unless you have to buy this book for a class, skip it (and if it is the text for a course, ask your instructor why!!)- buy Molecular and Cellular Physiology of Neurons by Fain instead, it doesn't cover as many topics, but breadth is no use if you can't understand the content. My department switched to the Fain text last year because so many people complained about Johnston et al.

    4 out of 5 stars I liked this book!.......2001-08-13

The book cover boasts that this is the only book with through discussions in mathematical equations, etc. Indeed! Except that there are other mathematically oriented books with kindly worked out examples. Some concepts, such as deriviation of the cable equation, were wonderfully presented. However, no attempt is made to obtain the solution. To think about it, that might have been an appropriate choice, for a physiologist does not have to know all the mathematics. However, the Rall model (Sec. 3.5) should have been approached in analogy with impedance matching. A unique feature of this book was somewhat detailed discussion of the Stochastic nature of ion channels. This is a good book, I believe. Overall, mathematics is thorough and abstract concepts are well chewed over.

2 out of 5 stars Dense and confusing.......2001-01-03

This book covers a lot of material. The quality varies from chapter to chapter. Figures are sprinkled here and there with very little explanation of what they mean. Additionally, there is not much consistency between figures. This book might be serve as a good companion to a course, but it's virtually useless for self study.

My background is engineering and neuroscience, but I still found the math to be poorly explained. If you're trying to learn about biophysics and cellular neurophysiology, pick another book. If you're unfortunate enough to have this book assigned to you for a class, make sure you attend the lectures.

The index is also terrible and virtually useless for anything that I've tried to look up. The only good thing about the book is that it references a lot of stuff, so you know where to look.

One star is for the breadth of topics covered and one is for the references. The Matisse drawing on the cover is also a nice touch.

4 out of 5 stars Understand the biophysics of excitable cells.......1997-12-16

You don't really have to hold a degree in mathematics or physics to understand cellular biophysics quantitavely; at least not with this book.

Clearly written, concise, self-contained, begins with the very basic knowledge of ion transport through semi-permeable membranes, moves to electrical properties of membranes, and then to passive and active properties of neuronal membranes, all in a very didactic and intuitive approach.

Of special interest are the chapters on stochastic analysis and formulation of single channel function, and on the relationship between calcium dynamics and transmitter release.

This text is essential for students interested in pursuing graduate studies in electrophysiology, cellular neuroscience, and in computational neuroscience.

          Book Description

Ion channels are crucial components of living cells. Situated in the cell membrane, they allow ions to pass from one side of the membrane to the other. This book provides an overview of ion channel study, including up-to-date coverage of the permeability and selectivity of channels, their gating and modulation, their response to drugs and toxins, and the human diseases that result from channel malfunction. Students of molecular biology, biochemistry, physiology, and neurobiology will benefit from this modern, concise text.

          4 out of 5 stars Excellent introduction.......2005-03-15

Getting a bit old now, but although it doesn't cover some of the most recent work it is by no means out of date. It remains one of the best introductions to voltage and ligand-gated ion channels and to electrophysiological methods of study. A good entry to the field, with an excellent bibliography of the primary literature. I don't know if I can recommend it at its list price (it is a slim volume), but used it could be a great buy.

            Book Description

Filling the gap created over the past five years, during which many new techniques have entered the market, this book is directed at both the new and the experienced ion channel researcher wishing to learn more about the considerations and methods for studying recombinant ion channels.

These latest developments are covered here for the first time, contributed by editors and authors working for major pharmaceutical companies and who routinely apply these techniques in their daily work. The first three chapters cover the use of the Xenopus oocyte expression system for structure-function studies, from basic approaches for manipulating ion channel cDNAs to more specialized but powerful techniques. This is followed by reviews of strategies and methodologies available for expressing channels in mammalian cells and for their analysis by patch-clamp electrophysiology. Chapters 6 to 8 review the latest methodologies for ion channel drug discovery, including high throughput screening using fluorescence and luminescence, as well as automated planar array electrophysiology. The remaining two chapters focus on approaches for determining ion channel crystal structures and on computational approaches to understanding channel mechanisms at atomic resolution.

Rather than provide detailed protocols, indicated by references in each chapter, the authors provide a comprehensive and easily accessible overview of the techniques involved, reviewing underlying principles and providing working guidelines as well as an understanding of the key theoretical and practical considerations associated with each topic. In each case, this practical advice is illustrated by real life examples, taken either from the author's own experience or from key examples in the literature, providing valuable practical hints not found elsewhere.

The result is a compendium of practical ion channel information that will prove a valuable resource to academic and industrial workers alike.
