          Book Description

Modern DNA microarray technologies have evolved over the past 25 years to the point where it is now possible to take many million measurements from a single experiment. These two volumes, Parts A & B in the Methods in Enzymology series provide methods that will shepard any molecular biologist through the process of planning, performing, and publishing microarray results.

Part A starts with an overview of a number of microarray platforms, both commercial and academically produced and includes wet bench protocols for performing traditional expression analysis and derivative techniques such as detection of transcription factor occupancy and chromatin status. Wet-bench protocols and troubleshooting techniques continue into Part B. These techniques are well rooted in traditional molecular biology and while they require traditional care, a researcher that can reproducibly generate beautiful Northern or Southern blots should have no difficulty generating beautiful array hybridizations.

Data management is a more recent problem for most biologists. The bulk of Part B provides a range of techniques for data handling. This includes critical issues, from normalization within and between arrays, to uploading your results to the public repositories for array data, and how to integrate data from multiple sources. There are chapters in Part B for both the debutant and the expert bioinformatician.

          · Provides an overview of platforms
          · Includes experimental design and wet bench protocols
          · Presents statistical and data analysis methods, array databases, data visualization and meta analysis

            Book Description

Research is fast paced and advances in RNA interference have recently opened up new opportunities for genetic experiments in human cell lines. However, the possibility to easily modify the genome still remains a powerful tool to investigate the function of coding and regulatory sequences in the vertebrate genome. DT40 has never been a quick and easy road to fame. It would be unfair to blame this on DT40 as it has proven to be a reliable and robust companion with fast doubling time, easy clonability and a relatively stable karyotype. If this model system is going to flourish over the next 15 years, it will be thanks to ingenious and original researchers. They may feel as if they work outside the mainstream, but they can take heart by the fact that only the clever exploitation of diversity and conservation makes biological research both elegant and rewarding. It is with this in mind that the DT40 handbook has been conceived and written. This book provides an up to date overview of the different facets of research, and also intends to help newcomers get started and avoid looming pitfalls. The collection of protocols which have been kindly provided by a number of laboratories will be particularly useful in this regard.

Serial Analysis of Gene Expression: Methods and Protocols facilitates the introduction of SAGE into the laboratory, and provides a framework for interpreting and comparing data derived from SAGE experiments. Of the several methods of genetic profiling available, only SAGE measures the expression of both known and unknown genes. SAGE studies encompass 50,000 tags and can provide detailed knowledge of the 2000 most highly expressed genes in the tissue sample. The SAGE protocols presented are detailed, fully annotated, and tested, and are all written by experienced SAGE researchers from around the world. Part 1 is dedicated to experimental procedures of SAGE and related methods including aRNA LongSAGE, SuperSAGE, DeepSAGE, and GMAT. Part 2 provides methods for extraction and filtration of tags, analysis of ditag populations, and completing statistically correct comparisons of gene expression profiles.

                Customer Reviews:

5 out of 5 stars I use this more than the actual book.......2007-10-08

I have both workbook and book. I use this when I'm plotting or replotting a book. I find the exercises make me think hard about my stories and bring out my creativity. I'd definitely recommend it.

                4 out of 5 stars Helpful!.......2007-08-24

I finally bought this book after rechecking it from the library several times. I love the challanges presented and the advice given. This book has helped me dig deeper and develop my own novel.

4 out of 5 stars Not the same as the Breakout book.......2007-03-14

Since he published Writing the Breakout Novel in 2001, Maass has taught a seminar of the same name at many writers conferences, and naturally learned from these experiences. The original book is here expanded by nearly three dozen exercises, which seem quite sensible, not hokey as are some by other teachers I've read.

I own the Breakout book and just now am comparing the workbook that I checked out of the local library. They're not the same, not by a long shot, despite what one reviewer here says. Though many sections have identical headers (such as Inner Conflict), they are completely rewritten, using different examples. The original book is 259 pages of 6x9, the workbook is 230 pp. of 8-1/2x11 format, thus it is by no means a condensation as TheCafeWriter asserts, and the original book is not necessarily more "in-depth." Some sections are, some aren't. The structure is substantially rearranged. Yet the concepts and the really fundamental points -- keep your story charged with tension, and so on -- do remain identical. These are essentially two complementary treatments of the same material by the same author

Maass asks his seminar participants to bring their in-process novels to perform exercises on, so the workbook is particularly useful if you are well into writing a novel already. With the discipline of the exercises, Maass teaches you to be your own draft doctor.

For me, there appear to be easily enough new perspectives and ideas here to warrant buying the workbook even though I have read the original and have it on my bookshelf.

5 out of 5 stars Good Hands On Exercises.......2006-08-14

This book is most beneficial to writers who have already completed the first drafts of their manuscripts. The exercises made me review some of my characters' attributes and turn offs as well as making sure my secondary plots tied into my main plot. Mr. Maass' suggestions caused me to re-think my villians a little bit and show a side to them that could be somewhat empathetic. Also, adding tension on every page for me was tough, but I found creative ways to do it. I do believe my completed novel is better now after making suggested changes. I stuck to my own gut on some of the things he suggested I change, but overall I did use a lot of his advice. This is a good purchase but you must do the written exercises not just skim over them.

                3 out of 5 stars It's good, but..........2006-04-01

Maass has created a good workbook with some nifty ways to come up with information. He's also included some material that's new, especially in terms of how to put together plots and subplots, and he expands the theme section from Writing the Breakout Novel.

Two problems though: first, except for a few places, the workbook really retreads the original book, just barely expanding on the original. I was hoping for more nuggets of information.

The second problem is that it is a workbook. I hate workbooks, mostly because my brain simply doesn't do those kinds of questions. I work from story down, not parts of story up. The workbook gives the impression to writers that answering the questions will magically make the story fall into place, and I instinctively reject that attitude. No, Maass isn't deliberately trying to tell writers the process, but the impression is there.

I would have rather he put together a follow-up book and then listed these questions at the end of the chapters instead of formatting the book with lines to be filled in. I think I would have been more receptive to the content. As it was, I read hastily through, feeling nauseous that I wasn't doing my homework--a kid's attitude from far too many workbooks in grade school.

Basically, good but not great information, and formatting that puts me off means that I have a hard time recommending this book. But if you're a workbook type, go for it.

                Book Description

This book will help you take a badly wounded patient to the operating room, organize yourself and your team, do battle with some vicious injuries, and come out with the best possible result. It is a practical guide to operative trauma surgery for residents and registrars, for general surgeons with an interest in trauma, and for isolated surgeons operating on wounded patients in military, rural, or humanitarian settings. A surgical atlas may show you what to do with your hands - but not how to think, plan, and improvise. Here you will find practical advice on how to use your head as well as your hands when operating on a massively bleeding trauma patient.

This book, subtitled "The art & craft of trauma surgery" is a gem. Worth every penny (or whatever coins you spend). A very practical book on how to handle major trauma of all organ systems, which first directs your thoughts and then your hands.

I thoroughly enjoyed this book not for the "how to do it"aspects of the book but for the "why to do it this way at this point in time" aspect. As the authors point out, much of trauma surgery is time critical, and options are often limited. Surgical decision making at this juncture is the factor that often leads to success or failure - Hirshberg and Mattox succinctly list options and the reasons for following one line of attack instead of another given different circumstances. The book is almost flippant in its language on many occasions, and for those of us who have done time at Ben Taub know that this is not in character for either author, for whom the art and craft of surgery is deadly serious. The apparent flippant tone introduced into the book allows the authors to provide emphasis in a way that a chart or graph wouldn't - fluently and with humour. The book is presented in the way that a mentor may review a case in the surgeons lounge - simply, clearly, and with relevance. This is an extremely useful book for those involved with trauma surgery, and I hope to see new editions of this book at regular intervals.

Um bom trabalho, livro de fácil entendimento e leitura. Lamento apenas a falta de detalhamento em vias aéreas.

overall a good book, with many practical tips. illustations are also good. i found the colloquial style irritating. ie. "The distal pancreas can be easily resected, while the pancreatic head requires a very big whack." These sort of practical manuals are valuable to practicing surgeons who know how to operate but can improve outcomes with the thinking portion of the operation, the art and craft of surgery.

Truly outstanding book for anyone providing surgical care to trauma patients. We've decided to purchase a copy for each of our senior residents so they will definitely read it. Easy to read. Practical knowledge and techniques, evidence based where possible, experienced base always. It reinforced much I've learned in 25 years of trying to teach trauma surgery and taught me new tricks and concepts.
