    Book Description

    As genomic science and potential gene therapies continue to develop, the detection of mutations plays an increasingly central role in diverse areas of biology. With innumerable detection techniques available spanning multiple disciplines, the choice of the best method for a particular study can prove exceedingly difficult. Complexity, equipment requirements, cost, and throughput are all factors to consider, along with the huge amount of information already available through genome sequencing projects.
    An essential go-to for informed decision-making, the Guide to Mutation Detection is the only single source for the theory and practice of mutation analysis needed to design an experiment. User-friendly, concise, and expertly edited by recognized leaders in the field, the text provides a cutting-edge reference that is still accessible to those with no experience in mutation analysis.
    The coverage is divided into two sections, with the first section reviewing such topics as:
    * Key technologies
    * Mutation scanning
    * Cleavage
    * Quality control
    * High throughput approaches
    * Databases
    * Nomenclatures
    The second section focuses on step-by-step protocols and discussion of methods, including:
    * Tag-array minisequencing
    * Electronic hybridization
    * Pyrosequencing
    * Fluorescent Single-Stranded Conformation Polymorphism (SSCP)
    * Denaturing High-Performance Liquid Chromatography (DHPLC)
    * Array Comparative Genomic Hybridization (ACGH)
    * Microplate Array Diagonal Gel Electrophoresis (MADGE)
    Throughout the text, the emphasis remains on practical applications. The detailedprotocols derive from the well-regarded Human Genome Organization (HUGO) training course, particularly those in Montpellier, France, 2002, organized by Mirielle Claustres, Johan den Dunnen, and Graham Taylor. Useful appendices contain important safety data, supplier lists, and bioinformatics resources.
    Up-to-date, user friendly, and comprehensive, the Guide to Mutation Detection is an invaluable resource for students and practitioners working across a wide range of genetics-related disciplines.

        Book Description

        This pathfinding book thoroughly identifies and explores the legal and ethical dimensions of the new genetics and the Human Genome Project. While accepting the goals of modern genetics research, the book takes a critical approach to the Human Genome Project. More precise genetic information will likely result in genetic discrimination, and will threaten genetic privacy in employment and insurance. The way we view disease, normalcy and our own humanness will undergo significant change as clinical medicine becomes increasingly genetics-based. We are more than our genes, but mapping them will move society into "genetics territory," and we may act as if our genes determine our destiny. This timely work brings together a group of the nation's leading experts in genetics, medicine, history of science, health law, philosophy of science, and medical ethics to assess the current state of modern human genetics, and to begin to chart the legal and ethical guidelines needed to prevent the misuse of human genetics from leading to the abuse of human beings. The six sections of the book, read together, map the social policy contours of modern human genetics. The first part describes the science of the Human Genome Project. The second addresses specific social policy implications, including the relevance of the recombinant DNA history, the eugenics legacy, military applications, and issues of race and class in the context of genetic discrimination. Broader philosophical issues, including reductionism and determinism, the concept of disease, and using germline gene therapy to "improve" human beings are discussed in the third part. The fourth is directed at the clinical implications of the new genetics, including privacy and confidentiality, genetic screening and counseling. The fifth part focuses on the legal and ethical frontiers in genetics, especially procreative liberty, patent issues, and regulatory mechanisms. Finally, a prioritized social policy research agenda is presented which suggests that immediate emphasis be on: 1) determining the rules for introducing a new genetic test into medical practice; 2) protecting the confidentiality and privacy of an individual's genetic formation; 3) preventing employment and insurance discrimination based on genetics; and 4) exploring how the new genetics will affect our concepts of "disease", "normalcy," and "humanness."

            Human Genome Methods is a practical guide to the application of molecular biology and genetics techniques to research on human cells. Written by recognized authorities who often originated the techniques described, chapters present experimental protocols that are readily used at the laboratory bench. The step-by-step protocols are concise and easy to follow to be reproducible by researchers of various levels of expertise. Suggestions for successful application of procedures are included, along with recommended materials and suppliers. Helpful background information and results of applying the methods described are also given. Section I covers topics such as microsatellite DNA, dynamic mutations, gene targeting using the DNA triple helix, and protease footprinting of DNA-protein interactions. This is followed in Section II by discussions of in situ hybridization, cell synchronization, and cell cycle specific gene expression. Methods concerned with programmed cell death are explored in Section III, which covers this emerging research area and the culture and analysis of cancer cells. Section IV presents methods related to transgene analysis of mouse embryonic stem cells, generation and knockout studies with null mutant mice, and mouse models for human disease. The final section reviews genome mapping, with an emphasis on the construction of linkage maps and on somatic cell hybrids for mapping disease genes.

                Book Description

                Douglas Robertson has taken his father's classic book, Survive the Savage Sea, as his starting point. On board their 43-foot schooner, LUCETTE, the Robertson family set sail from the south of England in January 1971 — in June 1972, LUCETTE was holed by killer whales and sank in the Pacific Ocean. Four adults and two children survived the next 38 days adrift, first in a rubber life raft, then crammed into a 9-foot fiberglass dinghy, before being rescued by a passing Japanese fishing vessel. This is the story of the 18-month voyage of the LUCETTE, across the Atlantic, around the Caribbean, through the Panama Canal and into the Pacific, and how the family survived their shipwreck. It is a vivid account of the delights and hardships, the excitements and the dangers, the emotional highs and lows experienced by the family both before and after the shipwreck. The author has drawn upon a wealth of other sources, including his own memories of a life-changing experience, to bring us this true story of adventure, of relationships strained to the bursting point, of conflict and resolution – ultimately a very human and humbling tale.

                5 out of 5 stars A gripping narrative of survival, and especially, the difficulties of getting along together in extremely cramped quarters .......2005-11-07

                The Last Voyage Of The Lucette is the complete and previously untold true story of an event first described by the author's father, Dougal Robertson, in "Survive the Savage Sea." Here, the son interweaves his own perspective - he was eighteen years old at the time - with the original narrative of a harrowing nautical voyage. In January 1971, the Robertson family set sail from England in an attempt to circumnavigate the globe; eighteen months later in the middle of the Pacific, their 43-foot schooner Lucette was holed by killer whales and sank. Four adults and two children survived for 38 days adrift in a survival raft, then a 9-foot dinghy, until a Japanese fishing vessel rescued them. A gripping narrative of survival, and especially, the difficulties of getting along together in extremely cramped quarters and desperate conditions. A handful of black-and-white and color photographs illustrate this amazing story of family togetherness under the most unusual and trying situations.

                5 out of 5 stars A page turner that fires the imagination .......2005-05-27

                This book had me totally absorbed from page 1. An extraordinary and remarkable true story of a familys' trip around the world and astounding tale of survival at sea after their schooner was attacked by a pod of Killer whales and sank, leaving the family of 6 adrift for 38 days without food or water. Eloquently written, it presents with clarity and coherence scenes and events that pluck at the heart strings in describing their ordeals. The author has managed to place the reader at the centre of the action as their nightmare unfolds. This book will make you smile and even cry as you share the emotional turmoil and unyielding dynamics of a family living in close confines aboard the yacht as well as when they were fighting for survival in their raft. Based on his father's classic book "Survive the Savage Sea" Douglas, a young adolescent boy who was forced to become a man overnight, has added almost 60% of new material including pictures, maps and illustrations to create a real masterpiece that has already been screened on the National Geogrphic Channel as a docu-film. Once started you wont put it down till your finished.

                5 out of 5 stars Inspirational.......2005-05-26

                An awe inspiring account of the family journey. Compelling reading for all! I couldn't put it down.

                2 out of 5 stars Misleading and Confusing.......2005-05-12

                I thought this was going to be a retelling of the story first told by Dougal Robertson in his excellent Survive the Savage Sea. But his son, Dougal, is incorporating most of the text from the earlier book, and is even writing from his father's point of view throughout this book. So 30 years later he's writing as his father to offer additional observations, which are now mixed in with his father's observations. I'm returning this book to Amazon.

                This book is somewhat unique, not just because it focuses on positive quotations, but also because the way the quotes are categorized and subcategorized into rather unique groups. Overall I would say the book is great, but in my opinion, the quote selection, choice of categories, and balance of quotations in categories could have been better (the author really went overboard in some places, particularly in quotes about prayers, which takes up page after page after page). If you want a book of positive / spiritual /wise quotes, I definitely recommend this book, but would suggest that you buy A Collection of Wisdom by Rodney Ohebsion and Treausry of Spiritual Wisdom by Ando Zubko first.

                "As a writer, I like to use quotations to illustrate themes or principles, such as the value of self-confidence or the importance of hard work. Unfortunately, in traditional compilations, finding just the right phrase to illustrate such themes can be very difficult." ~Leslie Ann Gibson

                As a casual quote collector, I take an interest in any and all books of quotations. I found over 22 quotes I wanted to collect. Leslie has organized the quotes in a rather comprehensive manner. These quotes are written by, for, or about women.

                If you are reading through this book, you will find some repeated quotes under a variety of headings. This becomes useful if you are adding quotes to your writing or to lecture notes. I only noticed because I read quote books from cover to cover in one sitting.

                This book is divided into three main sections:

                Finding Fulfillment
                Developing Positive Habits
                Achieving Your Goals

                You will then find quotes organized into themed sections which are again organized into groups. Yes, this is a very organized book.

                The content pages are helpful for finding just the quote you are looking for and the Index is helpful for looking up quotes by your favorite authors, philosophers, artists, leaders, actors and other famous personalities.

                I enjoyed finding quotes by Katharine Hepburn, Rose Franken, Elizabeth Bowen, Julia Child, Ingrid Bergman, Agnes Repplier, Lucy Stone and Eleanor Roosevelt.

                "You must do the thing you cannot do." ~Eleanor Roosevelt

                I also loved the quote by Louise Driscall: "Within your heart, keep one still, secret spot where dreams may go."

                Under "Finding Fulfillment" you will find quotes for welcoming joy into your life or discovering your strengths. Under "Discovering your Strengths" you will find quotes about doing what you love, believing in yourself and finding things to be proud of.

                "The Woman's Book of Positive Quotations" is a handy reference book for writers, speakers and those of us who love collecting quotes.

An uplifting joy to simply page through at random, and a veritable treasure trove of imminently practical advice ("The attributes of a great lady may still be found in the rule of the four S's: Sincerity, Simplicity, Sympathy, and Serenity.") The Woman's Book Of Positive Quotations" is whole heartedly recommended for self-help, self-improvement reading lists, as well as Women's Studies supplemental reference collections.

                5 out of 5 stars Attention Authors & Speakers: Buy this book now!.......2002-12-19

                As the author of a self-help/relationship book, I'm always looking out for new resources to improve my writing or develop ideas. My first book used "The New Beacon Book of Quotations For Women" as a primary resource and my next book will use this book extensively.

                As a man I find that women writers tend to be more colorful, personal, and subtle in their choice of words. That is why I prefer this type of book over a giant glossary of quotations.

                The best part of this book is the way the quotations are assembled around motivational-personal development themes. The choice of these themes shows a lot of enlightened thought and understanding by the editor, Leslie Ann Gibson. I've never seen such a high-quality collection of quotations around such well-chosen themes.

                The only thing that I miss is a more thorough biographical index. It's hard for me to write or speak about what someone says unless the audience (and myself) knows who it is. So unfortunately, I have to spend a lot of additional time researching.

                But all things considered this is a truly fine book that all serious writers and speakers should have in their personal libraries.

                5 out of 5 stars You go girl!.......2002-11-22

                Unlike the previous reviewer, I liked how some quotes appeared under different headings, making it easier to reference to drop into emails, cards, etc. A great compilation of affirming, positive quotes, and a nice gift for Mom.

                4 out of 5 stars A Wonderful Selection.......2002-11-09

                This book features wonderful quotes, many by well-known, contemporary sources. I would have given it a 5-star rating, except that some of the quotes are repeated several times throughout the book under different subject headings. Nonetheless, there still are an impressive number of unique quotes here--not just the usual selection, but some true rarities.

                This is a very excellent book. Buy it. Why? It will help you to impact people. It contains Hundreds of thought prevoking "Thoughts".

                From what I can understand right now, Leaders seem to have this unique way of allowing there followers to understand and take to heart wisdom that is passed their way. How?

                Have you ever noticed that Leaders impact always, always seems to come from a sentence or two? Even though, thoughts seem to just flow from them like rain from a cloud.

                This book is full of those impacting Thoughts that not only you can read, but you can use to impact others.

                Positive Thoughts from Martin Luther King to Hellen Keller.

                        I agree with the other reviewers. This book definitely lives up to its title. On every page is Scripture that is positive and uplifting. But something is missing...the rest of the story. I would argue that we cannot fully appreciate or even understand the positive if we do not understand the negative. God never overlooked the negative and never made things prettier than they were. (one example...the book of Judges, especially the last three chapters.) In fact, God has often used them for His greater purpose and glory. Those "negative passages" are there for a reason. And be that reason a lesson from the past, a warning for the future, or just plain hidden in the heart of the Lord, we should not go taking scissors to the Bible just to make ourselves feel better. Had this book merely advertised itself as "uplifting snippets from the Bible" it could have been a worthwhile book. But to raise itself up to equality with the full Bible, even infer that it is better because there is nothing negative? No, my advice...buy a full Bible and get the whole perspective and the whole message and the whole posititve "feeling" from knowing that God is both wrathful and loving, the Giver of grace and judgement, ever jealous for His glory and His people.

                            4 out of 5 stars Excellent review for Step 3.......2007-06-19

                            I'm finally finished with these stupid exams! Like probably all of you, I didn't have much time in between work to study for this exam. Now more than ever it was important to find something that provided the most information in the least amount of time.

                            First Aid was really helpful here. The High Yield facts was helpful and packed full of what you needed to know for the MCQ's on the exam. But the book really stands out with the 100 CCS cases. I read this section a couple of days before the exam, along with doing the USMLE CD. It was very helpful in providing the language necessary to complete the cases quickly. To my knowledge, no other written source provides this review.

                            Even though First Aid was not the only source I used to study for the exam, I think it's absolutely necessary for a quick read to pass the last of these painful exams!

                            I didnt have alot of time to study for the boards. This book was definatly a fast efficent review of the pertent topics. The CCS part of the book is the most valuable.

                            5 out of 5 stars Excellent Service My Friend.......2007-01-30

                            Got the book in a timely fashion, and in superb condition. Thanks so much! :)

                            3 out of 5 stars There has to be something better!!!!.......2006-12-16

                            I'm about to take my step 3 exam and I based my study on this book. It is divided in 2 sections, high yield facts and clinical cases simulator. The clinical case part is quite ok, gives you a rapid tool to do your cases. The high yield facts part, on the other hand, has several errors and facts that I really don't know where they came from and cost me some questions during questions practice... Since I started to doubt of the accuracy of this so called high yield facts, I had to invest more time in confirming some of them. There has to be a better book.
