                  In V(D)J recombination, the RAG proteins bind at a pair of signal sequences adjacent to the V, D, or J coding regions and cleave the DNA, resulting in two signal ends and two hairpinned coding ends. The two coding ends are joined to form a coding joint, and the two signal ends are joined to form a signal joint; this joining is done by the nonhomologous DNA end joining (NHEJ) pathway. A recombinational alternative in which a signal end is recombined with a coding end can also occur in a small percentage of the V(D)J recombination events in murine and human cells, and these are called hybrids (or hybrid joints). Two mechanisms have been proposed for the formation of these hybrids. One mechanism is via NHEJ, after initial cutting by RAGs. The second mechanism does not rely on NHEJ, but rather invokes that the RAGs can catalyze joining of the signal to the hairpinned coding end, by using the 3'OH of the signal end as a nucleophile to attack the phosphodiester bonds of the hairpinned coding end. In the present study, we addressed the question of which type of hybrid joining occurs in a physiological environment, where standard V(D)J recombination presumably occurs and normal RAG proteins are endogenously expressed. We find that all hybrids in vivo require DNA ligase IV in human cells, which is the final component of the NHEJ pathway. Hence, hybrid joints rely on NHEJ rather than on the RAG complex for joining. nd this point has implications for proposals invoking transposition to account for chromosomal translocations.

                    The vertebrate immune system generates double-strand DNA (dsDNA) breaks to generate the antigen receptor repertoire of lymphocytes. After those double-strand breaks have been created, the DNA joinings required to complete the process are carried out by the nonhomologous DNA end joining pathway, or NHEJ. The NHEJ pathway is present not only in lymphocytes, but in all eukaryotic cells ranging from yeast to humans. The NHEJ pathway is needed to repair these physiologic breaks, as well as challenging pathologic breaks that arise from ionizing radiation and oxidative damage to DNA.

                      I thought this book would be a technologically driven, marine-laden book about the sport. I gave it a try. What a surprise after only a few pages! Tim Ecott wrote a passionate book filled with historical chapters and memoirs of some of the best diving spots in the world, and he always includes the plants and animals as part of the experience. He introduced the underwater world to us landlubbers in such a way that the "underworld" becomes that which it truly is: another habitat that happens to be under water.

                      However, as the chapters rolled on, the book became a "history of diving" that showed the depth of research Mr Ecott did for this book. Jaques Cousteau and the Aqualung of the 1940s are what hastened SCUBA diving as we know it today; only the race to the moon is what slowed research down once the 1960s came.

                      This book is highly recommended to those spouses, friends and lovers who nag the SCUBA-obsessed divers. Give them this book and make them read it. They will then better understand the passion and beauty that's found only below the sea. Sea for yourself!

                      Understanding Neutral Buoyancy requires understanding the author. Tim Ecott is a reporter and producer for BBC World Service and has been writing for numerous prestigious international magazines and papers. He is a certified divemaster and a marine environmentalist. However, unlike the ultimate expert divers that have authored other diving books, Ecott is almost a reluctant diver. His father was a military man, and so young Ecott, a sickly, bronchitic child in his early childhood Wales fared much better in Malaya where his father was stationed for several years. A return to Ireland was a return to "varying shades of grey" for him. Though a lifelong swimmer, he came to scuba relatively late and his first experience was "just, well, fine." That daramtically changed later, but it's clear that this is a man who views diving as an emotional thing much more so than macho daring, socializing, or a scientific quest.

                      Tellingly, those who picked up Neutral Buoyancy with the anticipation of finding educational or instructional content regarding that important and celebrated aspect of diving will find it described in just one paragraph, an introduction to a chapter. The technical aspects are incidental; this book is really a collection of a wide variety of thoughts on diving, recorded by a deep and different soul, organized by an experienced journalist's mind, and crafted in exquisite language. Ecott, unlike many diving book autors, is a true writer, a professional, a master of language. Journalists and writers master the art of reporting facts and perhaps adapting them to the medium in which they will be published. In this instance, the medium is Ecott's own book where he is free to not just report, but also give his thoughts his personal spin.

                      Neutral Buoyancy is organized into a dozen chapters that each center on one general aspect of of things under the sea. There's, for example, a 30 page chapter entitled "Organic Gold" dedicated entirely to the sponge. Another deals with underwater habitats. There's "Flickering Images" that centers around Austrian diving pioneer Hans Hass and his wife Lotte, whom he seeks out and interviews. There's "Diving Free" that examines breathdiving record attempts and the whole experience around it. Or "In the Shadow of the Fire God" that describes a trip Ecott took to the Bismarck Sea. "Advanced French" deals with the various findings and advances a number of French pioneers brought to diving, most importantly, though not necessarily in Ecott's eyes, Jacques Cousteau (who he largely sees as a publicity grabbing egotist). There are other chapters dealing with underwater dangers, diving history, underwater warfare, pioneers, all presented in beautifully crafted language.

                      Ecott is a true citizen of the world. His world only, for sure, but of the world nonetheless. He travels to the places he seeks, delves deeply into them. His research is not just academic, no, he seeks out and interviews the pioneers, visits the places where things took place, and weaves it all into his words and descriptions.

                      Despite all this, I found the book an acquired taste. During the first half, I was often put off by what I found an overly negative view of things, one dismissive of essentially anything that wasn't old and untouched or at least made in the olden ways. I tired of the endless references on man's cruelty and thoughtless carelessness, and the somewhat manipulative hangdog way those thoughts were presented. That led me to becoming an overly critical reader who approached each new chapter with some bias, to the extent where I began faulting the writer for putting clearly British words like "programme" into an American's mouth. I actually put the book down for several weeks.

                      Then I picked it up again and I am glad I did. Having accepted Ecott's deeply personal view of the world and his tendency to craft personal biases into his accounts, I was finally able to appreciate the true magic of this book, the wealth of information and experiences it conveys without ever once falling prey to that old authors' vice, that of talking down or showing off, at least not in a technical sense. Neutral Buoyancy can be read and enjoyed by people who do not know diving, have never dived. His skillfully crafted brief explanations of diving basics explain without putting off experts while his gift of describing details, of truly painting with words, of conjuring up pictures and thoughts in a masterful way, will thrill even the most advanced diver.

                      I have just purchased my second copy of Neutral Buoyancy because I gave my first copy to the Manager of the Central Caribbean Marine Institute. This book is a must read for anyone who dives or is interested in the sea. Each chapter is prefaced by a dive story which either moved me to tears or laughter. I learned so much from Tim that I just had to get another copy of this wonderful book. Thank you Tim!

                      I absolutely loved this book! There are so many divers out there who try and "out macho" each other, so it was a really pleasant surprise to read a book by a person who just loves the sensations and and experiences of being underwater. I've said to a lot of people that diving keeps me sane, and I get funny looks; it was nice in this case to stumble across a somewhat kindred spirit.
                      I recommend this book wholeheartedly - it includes everything a reader could want - action, adventure, insight, and interesting and entertaining facts.

                      The Lonely Planet city guide to "Tasmania" is a very useful travel guide authored by Carolyn Bain, Gina Tsarouhas, and Paul Smitz. With 19 national parks which cover over a third of the territory, it will come as no surprise that a good portion of this guide is focused on outdoor activities. However, if you are headed to Tasmania for other reasons, as I am, there is still plenty of good information about sites to see, places to go, things to do, and places to eat. This review is based on the 4th edition of this particular travel guide, which was published in October of 2005.

                      The book opens with several sections about the state as a whole. There are sections on the history, culture, environment, the outdoors, national parks, and food & drink. It then follows with 12 more specific sections covering the major cities and different regional areas of the island, including a small section covering the Bass Strait Islands. The book then has a very useful section titled "Directory", which covers a large assortment of odds and ends which are useful for a traveler, such as business hours, climate, customs, embassies, festivals and events, holidays, insurance, money, and so on. There are then two more sections which cover transportation and health. The book then closes with a glossary, some information about the Lonely Planet books, a section for some notes, and an index.

                      I am fortunate enough to have friends there, so the main thing I used the guide for was to look at the descriptions of things to do, and to see what looks interesting. That blended well with my friends' suggestions, and I think it will prove to be useful to me in my trip. In particular, the "Directory" section is good to go through, as it can often serve as a checklist for things to think about and to take care of while you are planning your trip.

                        Having just got back from Tasmania with this book in tow I can highly recommend it to everyone, from locals, all Australians and the overseas visitor. This book has not only the history of this State but everything thant you could want to know about the flora and fauna, where to stay including the very basic accomodation through to the most luxurious, there are in depth maps, where to do the best shopping for your souveniers, where to eat whether it be fish and chips, vegetarian or a steak. This book was my bible and did not let me down once. We even found some short cuts with some lovely hidden treasures of nature displayed for us just by following the maps. The good thing was that the book does not only give information for the big towns but also many of the smaller ones hidden away,by visiting some of the smaller places It made my holiday that bit more personal and memorable. Read the book and you'll want to come and visit this wonderful isle.

                          I just got back on a trip from Tasmania where I used this book. Given that there aren't many tour books just on Tasmania, and the only other option would have been to carry around a huge Australia book with a little section on Tasmania, this was the best option. This not too shabby book proves there is enough to talk about in Tasmania, and so I think more tour book companies should publish single editions.

                          When I look at a tour book, especially Lonely Planet, I feel overwhelmed at all the information. And I think that's what LP does best - get the facts across. It's always good to have a second book about the same place for a second opinion. So as this book did fine at informing me, I also like to have a book that is more of an editorial, I don't think it's bad if in a tour book the author will give his/her personal opinion of something. This is what makes Rick Steve's so popular. Unfortunately he seems to only do Europe.

                          Rough Guides are good at giving opinions, and I think they are laid out in a much more accessible way. Also Fodor's just has one page where they list the big important "to do's" in one area; and I don't think that's a crime.

                          As a traveler I wasn't very selective because I went all over the island and saw just about everything.

                          So here's my little editorial:

                          Launceston: A nice town, smaller than Hobart. I think it's best to know someone here; otherwise I wouldn't spend much time.

                          Hobart: A great town, rich in history and activities. Try to be near the Salamanca boardwalk. The fantastic flee market happens there on Saturday mornings and it's also the general center of commotion. If you happen to be around during the "10 Days on the Island" festival, (early April) there will also be a lot happening there too.

                          Bruny Island: Skip it.

                          Tasman Peninsula: This large promontory sticks out of the south western part of the island, and along with great (bush) walking, also contains the islands premier tourist attraction, Port Arthur. This is a famous jail where the worst British convicts were sent. It has impressive buildings and a church, but probably wouldn't rank up with what you've seen if you have been to Europe. For history however, it is a great stop. I just went on the ghost tour, which was a waste of time. Go during the day.
                          Also in the Tasman Peninsula was the best hike I took on my stay. It was to Cape Raoul, just south of Port Arthur. Really impressive sea cliffs and you're looking straight at Antarctica from the view. You may want to spend one night on the peninsula, but keep in mind it's the countryside, so not the center of it all. The Comfort Inn is surprisingly nice, and has a good restaurant.

                          And of course, go to the devils park, seeing them fed is priceless.

                          West Coast: Strahan is a tourist rigged port, but still a trip up the Gordon River is worth it. Stay at the Gordon Gateway, eat at Risby Cove.

                          Cradle Mountain: A necessity, this is the big nature attraction in Tasmania. The walk around Dove Lake (which sits below the mountain) is unforgettable.

                          Mt. Field Park: Here I did the most strenuous hike, straight up to an alpine plateau. Great scenery but I wouldn't do it unless you really want your ass beaten.

                          Freycinet: Interesting mountains, and seeing Wineglass Beach makes you think you're in the tropics, wish it was warmer.

                          On a recent trip to Tasmania I refered often to this book and found it a useful companion. Like all Lonely Planet books it has good coverage of contexts such as political, historical, natural. There aren't too many guidebooks specifically devoted to Tasmania, and this one does the job admirably.

                            I still remember when I read this book in junior high school, and I'm getting ready to graduate this May from college, yet it still puts a smile on my face when I think about the book. It was one of those stories that you think could never happen to you, but it can, and it did happen to Melanie. I really enjoyed it, I suggest it to anyone looking for a lighthearted, good read. And for a little bit of a sequel, read "Eleven Kids, One Summer," which features Melanie, Justin and their families once again. But first, read "10 Kids No Pets."

                            This book was awesome. I never thought that Justin was a tv star. I was just as suprised as Melanie when I read the part about him being a star. It has all the things I'm looking for in a book. It has romance, twists, and it keeps you wanting to know more about their new found romance. I just wished it would have gave you a better picture of how their relationship worked out in the future. It needs a sequal.

                            I just read this book and i thought it was fantastic. I could relate to Melanie in so many ways. Justin is everything a guy should be because he was so sweet and kind and just a gentlemen. I knew something was odd with him going to "work" but i couldn't see it. Ann M. Martain it one of my favorite authors, so i knew this would be good. I hope theres a sequal coming.

                            I first read this book when i was in Junior High school in the 7th grade. I hated books, but one time i had to do a report in order to pass my class. The teacher gave us a few books and we had to pick one of them. I picked Just A Summer Romance because it looked intresting. I started to read the book, and from the first page i read i liked it. I liked it because it reflected my live. I loved that book so much that i read it in three days, i always read the book during math, spanish, social studies and english class, i use to wake up at three in the morning to read the book because i was wondering what would happen next. Now i'm a senior in High School and i still haven't forgotten about that love story.

                            I read this book when i was in 6th grade.i'am a junior in highschool now and still love the book.it really captures a true meaning of a first love.i thought the book was real romantic,and i thought justin was just a sweethart.melanie's character remided me of myself, of my first love.The book was so heart warming it was excelent it should become a movie i loved it.

                            Just for the Summer is aptly named, in that it is ideal reading for anyone slightly stunned by too much fun in the sun. Neither the story, the characters, nor the mystery itself will prove too taxing, but certainly Van Wormer's tale of sex, death, and money among Long Island's upper crust will provide hours of breezy entertainment for those long summer days.

                            I actually laughed in places when I read her stiff attempts at pretending to be of this century. I don't need a lot of sexy stuff to keep me hooked, just a decently plotted story (no go) and a main character I can care about. I would kill this girl if she were my houseguest. It's almost worth the slow read to see the word "golly" written about a gay liason and "down there" referencing, well...you know.

                            How did this book ever get published? It was the most boring thing that I've read this year. The entire book is about 474 pages. Just imagine wasting all that time for nothing. I believe that this novel should be considered general fiction and nothing more. There was some romance in it, but hardly any suspense. It just went on and on. The main characters went to about a thousand dinner parties and banquets. That took up about 75 pages in all! Nothing exciting happened at all until about page 250. But don't get me wrong, Laura Van Wormer is a good writer, because I'm reading "Talk" and loving it! Just trust me, save your money on "Just For The Summer."

                            I don't expect much in summer paperbacks, but I could not believe how this one dragged. How can someone take a story about murder, rich movie stars on vacation, and romance and end up with something so predictable, trite and boring? None of the characters were interesting or even distinguishable. Worse of all, the writing style was so dully flat that I felt like the author was whacking me on the head with each sentence. There is such a thing as excellent, entertaining trash that evokes emotion - Nora Roberts, Olivia Goldsmith, etc. The only reponse this book evoked was the realization that I wasted an afternoon.

                            Just for the Summer is a great read for the beach! Saturated with celebrities and juicy tidbits about the stars, this book was a tremendously fun read. I lost myself in the frolicking of the wealthy Hamptonites, putting the book down only to refresh my margarita!

                            The story was so in tune with the Hamptons, you find yourself in in the cottage, on the beach, also turning pages wanting to see what happens next.
