The condensed protocols version of Molecular Cloning is well written, concise and adequately referenced. Protocols are easy to follow and provide options depending upon individual experimental needs and preference.
The protocols provide information from home made recipes to prepared reagents available commercially.
The protocols work in my hands.
The book is large and not easily used on bench top. Otherwise it is a good resource for the laboratory.

This revised version of the Standard Lab Handbook has been improved, completed with new techniques and search is facilitated by the indices added at the end of each volume.

For many years the previous edition of this set was an essential reference in molecular biology labs. At present however, there are too many good protocol books out there to really make this argument. The book is pretty strong in explaining theory, and answering the question of why certain procedures are either necessary, useful, or worthless, however it is not as practical as many other books, such as Short Protocols. Still a good reference overall, but no longer stands alone, and I recommend checking out as much of the competition as possible before deciding whether to make the investment in it.

In this 3 volume set of books the authors summarize the most important laboratory protocols for DNA analysis and cloning. As someone involved in computational biology and mathematical gene sequence analysis, I was needing such a summary to get an idea of just how genetic engineering is actually practiced in the laboratory. The book is definitely written for those readers that are very experienced in these "wet" techniques, but it still could be perused profitably by anyone who is curious about genetic engineering. There is also an excellent website that owners of the books can go to and search for protocols and obtain updates and additions to the protocols.
At the beginning of each chapter, the authors give an introduction to the protocols and this is of an enormous help to those readers with only rudimentary acquaintance with the laboratory procedures. Typically, this introduction contains an historical summary of the procedures as they were developed or discovered. One can only marvel at the ingenuity of the discoverers of these techniques. These introductions are fairly straightforward to read, even for those that are not experts in biochemistry.
At the end of each chapter, the authors include an "information panel" that gives a more in-depth view of the biochemistry or genetics behind the procedures. These are summaries and are highly specialized, and are again meant for experienced readers. A very lengthy list of references is also included at the end of each chapter.
Becuase of the size of this collection, space here does not permit a detailed review, so I will list some of the areas that I thought were particularly interesting or well-written (these coming from the introduction or the information panels only): 1. The DNA synthesis at the colE1 replicon and the interaction between RNAI and RNAII. 2. The discussion of electroporation and the physics behind this technique to introduce DNA into eukaryotic cells. 3. The discussion on the discovery of bacteriophage lambda. 4. The discussion (with diagram), of the assembly pathway of bacteriophage lambda. 5. The summary of the early analysis of DNA using electrophoresis and the different pulsed-field configurations used. 6. The anecdote on the discovery of the polymerase chain reaction. 7. The short discussion on computer-assisted design of oligonucleotide primers. 8. The discussion of oligonucleotide synthesis. 9. The flowchart detailing the preparing and screening of a cDNA library. 10. The history of the development of the methods to synthesize and clone cDNAs. 11. The detailed discussion of the molecular cloning of double-stranded cDNA. 12. The discussion on the methods to validate clones of cDNA. 13. The discussion on magnetic beads for affinity purification. 14. The discussion on the history of DNA sequencing and the different techniques to accomplish it, particularly the information panel on automated DNA sequencing. 15. The discussion of the different types of mutagenesis and the different methods for accomplishing it. 18. The fascinating discussion of how to introduce cloned genes into mammalian cells. 19. The discussion on the steps involved in DNA footprinting. 20 The discussion on green flourescent protein and its use as a fusion tag. 21. The discussion on the use of surface plasmon resonance spectroscopy.

Very useful.......2002-11-14
This book is very much like its well known predecessor, the currently 3 volume Molecular Cloning Manual with which it shares an editor and a similar title (in fact, it could pass as volume 4).
Overall it gives a good coverage of spotted microarray technology starting from the preparation of probes and slides to sample preparation and hybridization. In addition to expression profiling areas covered include uses of microarrays for analysis of chromatin immunoprecipitation samples, DNA copy number determination, and detection of genetic polymorphisms (oligo arrays). Tissue-arrays and micro-dissection techniques for sample preparation are also described. The bioinformatics section is less extensive than the experimental parts, but the chapters on clustering, self organizing maps and databases serve as good introductions to these areas. I especially liked the image acquisition, normalization and quality control sections. These reviews presented their material in a very clear and sensible way.
Each chapter starts with a short summary of the relevant background. These introductions are very concise, straightforward, jargon-free and up to date in their references. They are written by leading experts of their fields. In addition, there are several "Information Panels" with similar qualities, covering somewhat more general background material.
This is a very nice looking book; it is a pleasure to look at and to hold. I am sure every lab that uses microarrays could benefit from it.

Revised, Updated, Expanded Second Edition.......2006-01-25
This second edition is much changed from the first edition of 2002. Most of the chapters in the earlier have been either extensively revised and brought up to date or replaced with new chapters (the old chapters are still available on a special web site).
The format of the book is the same with forty-six chapters written by the leading researchers in the field. The chapters are grouped into sections as follows:
1. Protein-Protein Interactions in Biological Context
2. Standard Technologies to Probe Protein Interactions
3. Biophysical Approaches to Probe Protein Interactions
4. Novel High-Throughput Approaches to Probe Protein Interactions
5. Interactions of Proteins and Peptides
6. InVivo Imaging of Protein Interactions
7. Genome-wide and Computer-based Analysis of Protein Interactions
8. Design of Novel Interactions and Inhibitors in Drug Discovery
This book is a summary of the state of the art in the field and is significantly improved over the first edition.
some good looking protocols - but bad mistakes: useless for a real scientist.......2005-09-27
Most of the protocols may be as good as they appear - no doubt about it! Most of the protocols have sure been written by well established scientists with a lot of input into details. But CSHLpress has an extremely good reputation to edit the very best books like the so-called "Maniatis" known for more than two decades as "the" standard in "molecular Cloning". With this respect the first edition of "Protein-protein interactions: A molecular cloning manual" only "copies" the famous word "cloning" on the title of the book and uses (legally) the logo "CSHLpress" to give the impression of a similar standard. But this impression is wrong: simple mistakes in the introduction of a method support strong doubts about how valuable the following protocol could be. This is not a good example of "CSHLpress"'s own extremely high standard. But hopefully most of the methods represent a good and detailed overview which may (or may not) work. I suggest to avoid the first edition and check carefully the 2nd edition soon. There is at least some room for improvement to reach the otherwise often unreached standards of CSHLpress.
simple mistakes - appears as no scientist ever checked for mistakes?.......2005-07-20
the very first impression of the book is pretty good: it appears to collect many useful and detailed protocols - useful especially for students and attractive for supervisors - and of course libraries. Unfortunately the first impression doesn't hold: very simple (and easily avoidable) mistakes show that neither the authors (or at least some of them) never ever checked for logistic mistakes - therefore it is very questionable if the protocols ever worked the way they were described. There is only little hope for an improvement of the (soon appearing) second editions. It may be worth for students to check the original literature where at least some peer review process could improve sophisticated protocols.
