A core book designed mainly for the nonspecialist, "Flow Cytometry: First Principles" is addressing in ten chapters the essential issues in flow-cytometry, from flow machines to reagents, techniques and their applications in fundamental research and clinical settings. The first chapters conveys us as back as the beginning of the century, when the concept of flow-cytometry germinated. Light microscopy, cell counters and ink jet technology are standing at the grounds of flow-cytometry and their merging paved the way toward the modern, "black box", flow cytometers. However, "beyond the black box", the instrumentation and the technical aspects are comprehensively described in several chapters. a discussion on data acquisition and analysis is provided, along with the most used cell staining procedures, fluorochromes, lenses, mirrors and lasers. A widespread application in flow-cytometry is cell labelling, particularly leukocyte subpopulations identification with monoclonal antibodies binding cell surface specific markers. Although lymphocytes are the prototype, methods presented here -dot-plot, histogram, quadrant analysis and gating are applicable to many other cell types , including bacteria or algae. With the advent of some "state of the art" machines, DNA and chromosome analyses become feasible by flow-cytometry. It is foreseeable that molecular biology on flow-cytometer will not stop to cell cycle, ploidy and apoptosis investigation, but even chromosomal translocations or point mutations would be detectable with enough precision for diagnosis. Beside fundamental research applications, flow-cytometry is now an indispensable technique in the clinical laboratory, too. Oncology, immunology, hematology and pathology are fields where flow diagnosis has sometimes the last word to say. Cross-match analysis and HLA phenotyping can now rapidly be performed by flow-cytometry, with usefulness in organ and bone-marrow transplantation. The final chapter is intended to present the flow-cytometry of the future, as seen in 1992, with an emphasis on the functional assays which are gaining now an increasing interest. However, a precedent, "out of the stream" chapter is preparing us on what this future could look alike. It approaches some of the border-line pplications, in fields where flow-cytometry is becoming a current tool of research. These are microbiology -prokaryotic DNA and drug resistance determination-, ecology -aquatic environment, plankton, algae-, developmental biology or the functional studies -Ca2+influx. The book is highly recommended for flow-cytometry and sorting beginners, but it is an equally pleasant lecture for people working in many life sciences fields, eager to find out how flow-cytometry is shaping current biological research.

    I took this guide to Europe recently. A lot of the recommendations were good, but the guide is not totally useful while on the trip. For instance, the maps that they provide always seemed to cutoff where I was staying, or don't include all of the train stations, so you don't know how to orient yourself without getting a better map. I realize that the maps focus on the more touristed parts of town, but they do not mark the recommendations on the map, so you're left with an address, and a crappy map that doesn't list streets very well. Rick Steves has the tourist area maps, but he marks his recommendations on his maps, making it much easier to actually use the guide. The Rough Guide makes a lot of good recommendations, it's not as quaint as Rick Steves, which was good for me, but Rick Steves guides are much more user friendly. Whenever we tried to find something using his guide we found it. With the Rough Guide we only found it by chance, or through looking into it on our own at a tourist offfice.

    I used the Rough Guide Europe as my guidebook for a three-month trip through twelve countries. There are seven advantages the Rough Guide has over its competitors:

    1) The breadth of coverage -- 32 countries with info on tons of cities and towns, reviews/descriptions of the sights, and options for hiking, cycling, paragliding, and other adventurous pursuits.

    2) Thorough listings and reviews for accommodation (hostels primarily, but also hotels and B&Bs), eating, drinking, and nightlife.

    3) It's small and weighs next-to nothing.

    4) The intuitive, aesthetically-pleasing, user-friendly layout makes it easy to find what you're looking for.

    5) Fantastic maps.

    6) Everyone else seems to use either Let's Go or Lonely Planet -- this is a tremendous advantage.

    7) Loaded with travel details that include basic timetables for trains, buses, and ferries.

    This guide book is designed for those who seek to get off the beaten path, and explore Europe on their own. Rough Guides have never let me down. More to the point, they have enriched my travels more than any other guidebook available.

    If you are an independent budget traveler planning on a trek to Europe, you would be hard-pressed to find a better guidebook.

    The Rough Guide to Europe is excellent in providing a lively overview of every country in Europe (plus Morocco). Rough Guide is unafraid to put in the good with the bad, giving a clear critical analysis of what's worth seeing and what isn't. All the cities and destinations one would expect to find--from Slovenian resorts to Paris to Santiago de Compostela--are included in here.

    Of course, putting all the relevant information for the whole of Europe would require several dozen 2000-page travel guides, so travelers to a specific country will also want to pick up one of Rough Guide's country-specific books. Yet if I had to backpack across the whole of Europe with a single book, this would be it!

    The maps for such cities as Sevilla, Venice, and Lisbon are excellent for the brief visitor, and really give a feel of how to distribute your all-too-often limited time.

    If there is any minor issue I have with this guide, it is that the hotel selections are limited and generally inferior to what you might find in a full guide to a particular city. Of course, this goes back to having to squeeze in several dozen nations in one book.

    All in all, if you are planning on visiting several countries or just meandering through Europe, Rough Guide to Europe is a one great buy.

    What I like best about the Rough Guides is that they give critical reviews. They will point out the tourist traps and will give negative reviews. I find that Frommer's and Fodor's rarely point out the negatives to a particular... The rough guide is very critical and is a great balance to these other guides.

    If you aren't interested in "roughing" it and staying in lower priced hotels.. the guides are still very useful in rating attractions, and areas in which to stay... but you will need another book to look at more moderate and luxury hotels.

    I would definitely read this book before going to Europe.

    The start of this book was a little rough for me. The author introduced all the character all at once and I was pretty confused for a while, but once you get into the story you're hooked! This book was like a great episode of Dateline or 20/20. It was full of excitement, mystery, and really kept you on the edge of your seat wondering what happened and what was going to happen. It would be great to take todays forensics to figure out some of the things they couldn't figure out back then...sadly most of the artifacts from the doomed Karluk expedition have been lost.

    Coincidentally I had just finished 'Alive' - another story about cold weather survival. Perhaps mainly by contrast I found it impossible to get into 'The Ice Master'. While the event is undeniably interesting, Niven's telling ruins it.

    I love reading the stories of the great arctic adventures...This one is awesome! It's basiclly the opposite of Shackeltons Endurance...No comradery, lots of death, a coward for a leader and even at the other end of the world. It really offers up a contrast. Yet through all the struggles heros still arise to meet the challenge.

    I absolutely could not put this book down. We had a house full of family guests and I found myself sneaking away just to try and grab a few more pages of this thrilling, yet horrifying account of the voyage of the Karluk. On one hand, I almost dreaded to pick the book up because Niven truly brings these characters to life and you become very involved with them. On the other, I just had to know who would and who would not survive and how the ones who did survive managed to do it. Niven did a superb job all around.

    For me "South", is the classic Polar expedition tragedy book. This book is on par with it. However in any second source history book, there are sometimes some niggling details that are wrong. The fact that another "doomed" expedition had some survivers, may not have been known to the crew on this one. But its not clear that is what happened. Now I want to go find that book and read it. But had you only read the original books & sources you would have come away with a completely different perspective. Ms. Niven got to the heart of the trip and who were really the heros here and who were the cads.

    But, this is more than just a man triumphs over nature book. Some of these folks survived but it wasn't great. It was a good read though and I'm glad I picked it up. I'm going after Ms. Niven's other book "Ada" next.

    This true story of survival has been the subject of two other books and a movie. In 1994, a ten-man group of soldiers, 7 British and 3 Chinese from Hong Kong, went to Sabah, Malaysia with the intention of being the first ever to successfully navigate Lows Gully. Lows Gully is a deep chasm off the northern flank of Mount Kinabalu. Superstition, mystery and intrigue surrounds this place instilling fear into some of the locals. It was into this situation that these men went to create history.

    The other two books that were written about this expedition came from four of the team involved. With regard to this book, R.M. Connaughton is independent. He seems well qualified to undertake such a task having a military background and actually serving in the Far East. At the beginning of the book, he provides historical, yet essential background information to Mount Kinabalu and Lows Gully. He relates the exploits of Hugh Low (whom Lows Gully was named after) and Spenser St John. In the 1990's the attempt by Robert New and Steve Pinfield is also related.

    With the background information, Connaughton then begins to describe the preparations for the expedition describing the building of the team and the travelling to Mount Kinabalu. Their is a photo section in the middle of the book and at the end there is a chapter entitled "Reflections" which describes some of the findings of the subsequent Board of Enquiry.

    The book highlights the various tensions that existed between team members and tries to establish what exactly went wrong. Connaughton does an excellent job with this difficult task. The chapter detailling the extensive rescue operation that took place with the British and Malaysians deserves a special mention.

    This is an excellent book and you can almost envisage the savage jungle terrain that is described as you read it.

    On the whole this is a well-written and well researched book and I thoroughly recommend it.
