Book Description

Few technical manuals have become standards in biomedicine. Antibodies: A Laboratory Manual, by Ed Harlow and David Lane, has had that status for a decade. Now there is a new and even higher standard—Using Antibodies: A Laboratory Manual.

Harlow and Lane have completely revised their guide to the use of immunoglobulin reagents in the laboratory. Chapters have been entirely rewritten, reorganized, and updated to provide background, context, and step-by-step instructions for techniques that range from choosing the right antibody and handling it correctly, to the proper methods for characterizing antigens in cells and solutions. New chapters on tagging proteins and epitope mapping are included.

Rather than presenting an array of solutions for working with antibodies and antigens, Using Antibodies instead identifies in each case the best approach to specific problems. These recommendations include more detail in the protocols, extensive advice on avoiding and solving problems, information regarding proper controls, and extensive illustration of theory, methods, and results, both good and bad. An additional bonus included with this manual is a set of Portable Protocols, step-by-step instructions for the most frequently used and essential techniques printed on spill-proof, durable cards that can be annotated and used directly at the bench.

The expert advice in Using Antibodies is presented using an imaginative design with extensive use of color and graphic elements calculated to help readers plan and execute their experiments efficiently and accurately. A newly available type of binding will maintain the manual's integrity during years of use.

1 out of 5 stars Should be [less money], Not [more money]!.......2002-10-21

Although I liked the outdated 1988 version of this book, this one is extremely simplistic and omits much of the previously included material. Most of the information included can be found on the web and is shallow compared to Current Protocols or other texts. Although it serves as a reasonable introduction for novices to this area it's extremely overpriced for [money]!!!

I would reccomend Monoclonal Antibodies by Goding ([money]),

5 out of 5 stars Two Thumbs Up.......2000-10-25

This is an exceptional reference for anyone using abtibodies in the laboratory. It is a *must-have* for anyone venturing into unknown teritory. For the experienced individual forced to bang out new protocols with a cranky antibody, this reference is priceless.

3 out of 5 stars Good manual, though needs revision........2007-06-28

This laboratory manual is very concise and easy to read. The topics range from antibody production to protein/enzyme conjugation, with step by step instructions and formulations. Previous to most topics is given a fairly thorough background. One drawback is the fact that this book is nearly 20 years old (pub. 1988) and lacks a significant amount of progress made in the areas of homo and heterobifunctional linkers, PEGylation, and antibody engineering. Given a good price however, this would be a good reference manual.

4 out of 5 stars An old classic, but still useful........2002-08-26

    This classic is getting old...
But you cand find some important informations inside, classic protocols and keys-data to the antibodies-world.
A must-have reference for any today-biologist working with antibodies (immunostaining, immunoprecipitation, Western-blot, antibodies production, etc...).

4 out of 5 stars An excellent summary of immunological methods........1998-01-16

This book is indespensible for those wishing to set up any type of antibody related methodology. Gives excellent overviews of immunodiagnostic methods (ELISA,RIA,etc.), antibody production (polyclonal and monoclonal), and includes basic summaries on the nature of the immune sytem in vivo. Especially useful to researchers developing immuno-techniques as it gives detailed methodologies.

        Book Description

Prion diseases, also known as Transmissible Spongiform Encephalopathies (TSEs), exist in both humans (Creutzfeldt-Jakob disease (CJD)) and animals (scrapie, bovine spongiform encephalopathy (BSE), chronic wasting disease) and have the unique property of being infectious, sporadic or genetic in origin.

Although the precise nature of the infectious agent responsible for TSEs is not definitely identified, it is now clearly demonstrated that a protein named PrP (for Prion Protein) plays a critical role in the transmission and pathogenesis of TSEs.

This book provides the general description as well as the details of the techniques currently used for the study of prion diseases. Taking into account the pivotal role played by PrP it is not surprising that many Chapters of this book deal with the purification, the detection and the characterization of the different forms of this protein. In addition, in vitro, cellular and animal models specifically adapted to the study of TSEs, as well as bio-safety procedures are described. Each Chapter is written by scientists involved for many years in their respective domain of prion biology who give the best of their knowledge in this technical document. This volume is a very useful tool for any laboratory which recently decided to contribute to the study of TSEs as well as for teams already engaged in this field for many years but interested in extending their technical capacity toward new methods.

        Features:

Purification of PrPC and the Pathological Isoform of Prion Protein (PrPsc or PrP-res)

        Animal Models of TSEs

        Cell Culture Models of TSEs

        Western Immunoblotting Techniques

        Antibody Production and ELISA

        TSE Strain Typing in Mice

        Biosafety and Decontamination Procedures

        Cell-free Conversion of Prion Proteins

        Cytotoxicity of PrP Peptides

Cyclic Amplification of Prion Protein Misfolding

        Of interest to:

          Book Description

Polyclonal antibodies, widely used in research and diagnostics, are conventionally isolated from the blood of immunised mammals. The fact that antibodies can also be detected in the yolk of eggs laid by immunised hens, led to the development of the yolk antibody technology as an alternative method less stressful to animals. Hens can be kept under nearly natural conditions and antibodies be isolated from the eggs. Further, the IgY-technology has the advantage that avian antibodies can be produced against several antigens which do not induce a immune response in mammals. Protocols on how to keep and immunise hens and on the extraction, isolation and use of antibodies from yolk are described in detail in this manual. For their work on yolk antibodies the authors received the FISEA (International Foundation for the Substitution of Animal Experimentation, Luxembourg) prize in 1997.
